Introduction

Cellular interactions with the extracellular matrix (ECM) play key roles in the assembly of cells into multicellular organisms and profoundly influence a variety of signalling events including those involved in survival, proliferation, differentiation and migration. Such cell-matrix contacts take place in physiological situations (ie development, immune response, tissue repair…), but also in pathological events (ie cancer, inflammation…). Adhesive interactions are mediated by a network of ‘anchor proteins’, some of which directly mediate the linkage between the actin cytoskeleton and the membrane, whereas others play a regulatory role. Whereas integrins are the major cell surface receptors for the ECM, other adhesive systems have also been described, including matrix and transmembrane glycosaminoglycans (GAGs) molecules such as syndecans. Both the expression pattern of these ECM receptors and the ligand-binding affinity determine the adhesive properties of the cell. The ligand-binding affinity may be regulated by intracellular cues, but through extracellular interactions, adhesion receptors are also capable of signalling to the cellular machinery and thereby affecting gene expression and several cellular functions. Our group studies cell-ECM interactions during tissue remodelling such as wound repair and epithelial tumour progression. The accomplishment of our research project relied on two major strategies: (1) one facet is dedicated to the analysis of the molecular mechanisms underlying cell-ECM interactions at the fundamental level and (2) the other aspect of our work consists in taking advantage of our expertise in the field of cell-ECM interactions to develop strategies aimed at tissue reconstruction. We focused on skin and cornea repair.
