Bien que ces stress diff\u00e8rent par leur nature et leur intensit\u00e9, ils sont g\u00e9n\u00e9ralement int\u00e9gr\u00e9s par des r\u00e9seaux de signalisation mol\u00e9culaire qui mobilisent des capteurs et effecteurs communs. Cette int\u00e9gration aboutit \u00e0 l\u2019activation de programmes adaptatifs complexes visant \u00e0 restaurer l\u2019hom\u00e9ostasie cellulaire ou, \u00e0 d\u00e9faut, \u00e0 engager des voies alternatives, telles que la s\u00e9nescence, la plasticit\u00e9 ph\u00e9notypique ou la mort cellulaire. Ces r\u00e9ponses conditionnent non seulement le devenir individuel de la cellule, mais influencent \u00e9galement l\u2019architecture et la dynamique des tissus environnants. Cette modulation du microenvironnement peut, dans certains contextes, contribuer \u00e0 la d\u00e9r\u00e9gulation des \u00e9quilibres tissulaires et favoriser l\u2019initiation ou la progression de pathologies inflammatoires chroniques et malignes, menant \u00e0 des stades cliniquement irr\u00e9versibles.<\/p><p style=\"font-weight: 400;\">Notre \u00e9quipe \u00e9tudie les m\u00e9canismes mol\u00e9culaires d\u00e9tectant et int\u00e9grant les signaux de stress microenvironnementaux alt\u00e9rant l\u2019hom\u00e9ostasie des prot\u00e9ines et des ARN. Ces perturbations, int\u00e9gr\u00e9es notamment au niveau du ribosome en cours d\u2019\u00e9longation, d\u00e9clenchent des voies de signalisation adaptatives (telles que GCN2 ou la r\u00e9ponse au stress ribotoxique) qui orientent les trajectoires cellulaires et favorisent l\u2019h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 ph\u00e9notypique. Nous analysons la dynamique transcriptionnelle et traductionnelle de ces r\u00e9ponses en conditions physiologiques, o\u00f9 elles soutiennent l\u2019hom\u00e9ostasie tissulaire, et pathologiques, o\u00f9 leur d\u00e9r\u00e9gulation promeut plasticit\u00e9 adaptative et r\u00e9sistance th\u00e9rapeutique.
